Everything else comes from the published label for that drug, which is where the half-life used here is taken from. The maths runs on your device as you answer, which is why the chart redraws the moment you change an answer.

The model treats your body as one space that the drug enters and leaves at a steady proportional rate. Each dose adds to whatever is still there from the previous ones, which is how a weekly injection builds a level over the first weeks rather than starting from scratch every Sunday. It is the same arithmetic used to explain half-life in a pharmacology textbook, applied to your actual schedule.

What it deliberately does not do is model you. It has no idea about your weight, your kidney function, what you ate, or how fast you personally clear the drug, and it is not fed by any blood test. Two people entering the same answers get the same chart.

What does this calculator actually show?

It shows an estimated medication level over time, based on your dose schedule and the half-life published on the drug label. The line rises after a dose, peaks, then falls until the next one. It is a picture of the pattern, not a reading from your body.

Is this the same as a blood test?

No. A blood test measures what is in you right now. This is arithmetic applied to a published average, so it cannot know your absorption, your kidney function or anything else specific to you. Treat it as educational.

What is a half-life?

The half-life is the time it takes for half of the drug to leave your body. For semaglutide it is about 7 days, so roughly half of a dose is still there a week later. That single number is what gives each medication its shape on the chart.

How long does semaglutide stay in your system?

With a half-life of about 7 days, about half remains after one week, about a quarter after two, and about an eighth after three. Most of a dose is gone after roughly five half-lives, which works out to about five weeks.

How long does tirzepatide stay in your system?

About 5 days. The Mounjaro label prints that figure and the overdosage section of the Zepbound label repeats it, while the Zepbound pharmacokinetics section gives about 5 to 6 days for the same molecule measured in adults with overweight or obesity. What is steady state and how long does it take to reach?

Steady state is the point where the amount you take each week matches the amount you clear each week, so the line stops drifting upward and just repeats. It takes about four to five half-lives, which is around four to five weeks for semaglutide and about four weeks for tirzepatide.

Why does the curve keep climbing when my dose has not changed?

Because a week is about one half-life for these drugs, so a new dose lands on top of a substantial leftover from the previous one. That stacking continues until clearance catches up with intake, which is exactly what steady state means. Nothing has gone wrong, and it is not a reason to change anything.

Why does the peak land a couple of days after the shot?

What happens to the curve when I move up to a higher dose?

The whole line shifts to a higher level and then climbs again for several weeks until it settles at a new steady pattern. The shape stays the same, the scale changes. When and whether to move up is entirely between you and your prescriber.

Do daily pills build a curve too?

Yes, and it is a much flatter one. Oral semaglutide (Rybelsus, Wegovy tablets) has the same 7 day half-life as the injection, but taking a smaller amount every day instead of one weekly dose smooths out the peaks and troughs. Orforglipron (Foundayo) has a published half-life given as a range of about 29 to 49 hours, roughly 1.2 to 2.0 days, so its line is drawn as a wider estimate.

Does liraglutide look different?

Very. Saxenda and Victoza have a half-life of about 13 hours, so the level rises and falls within a single day and almost nothing carries into the next week. A missed day shows up on the chart immediately, which is not the case for weekly medications.

Can I calculate retatrutide?

Only as a rough sketch. Retatrutide is not FDA approved and has no approved dose, and the figure of about 6 days comes from a phase 1b trial rather than a label. Anything drawn from a number like that should be read as provisional.

Does my weight, age or metabolism change the curve?

In your body, almost certainly. In this model, no. It uses population figures from published labels and applies them the same way for everyone, which is the honest limitation of any calculator that does not draw your blood.

Does the injection site change the curve?

Why is the result a percentage instead of a concentration?

Because a percentage is the part that is actually meaningful here. The chart compares today against the steady peak of your own dose, which shows you where you sit in the week. A concentration figure would look precise and would not be, since the model has no measurement of you.
